RESUMO

The energetic particle radiation environment on the International Space Station (ISS) includes both charged and neutral particles. Here, we make use of the unique capabilities of the Radiation Assessment Detector (ISS-RAD) to measure both of these components simultaneously. The Charged Particle Detector (CPD) is, despite its name, capable of measuring neutrons in the energy range from about 4 MeV to a few hundred MeV. Combined with data from the Fast Neutron Detector (FND) in the 0.2 to 8 MeV range, we present the first broad-spectrum measurements of the neutron environments in various locations within the ISS since an early Bonner-Ball experiment that was conducted before the Station was fully constructed. The data presented here span the time period from February 2016 to February 2022. In addition to presenting broad-spectrum neutron fluence measurements, we show correlations of the measured neutron dose equivalent with charged-particle dose rates. The ratio of charged-particle dose to neutron dose equivalent is found to be relatively stable within the ISS.

RESUMO

The spatial distributions of antibonding π^{*} and σ^{*} states in epitaxial graphene multilayers are mapped using electron energy-loss spectroscopy in a scanning transmission electron microscope. Inelastic channeling simulations validate the interpretation of the spatially resolved signals in terms of electronic orbitals, and demonstrate the crucial effect of the material thickness on the experimental capability to resolve the distribution of unoccupied states. This work illustrates the current potential of core-level electron energy-loss spectroscopy towards the direct visualization of electronic orbitals in a wide range of materials, of huge interest to better understand chemical bonding among many other properties at interfaces and defects in solids.

Leptospirosis is one of the most widely distributed zoonosis in the world. Bovine leptospirosis is a serious problem in bovine production, causing reproductive losses. The aim of this work was to compare recombinant LipL32 with sonicated antigen for detecting anti-Leptospira IgG antibodies in bovine serum using ELISA. The Microscopic Agglutination Test (MAT) is used as the gold standard. Sonicated antigen from cultures of Leptospira interrogans serogroup Icterohaemorrhagiae serovar copenhageni (strain M20) was used for the eELISA and rLipL32 for the rELISA. The performance of these assays was evaluated using serum samples from 166 bovines, 69 MAT positive and 97 MAT negative. At the optimal cut-off point recommended by the receiver operating characteristic (ROC) curve analysis, the sensitivity and specificity values were 98.6% and 97.9%, respectively, for eELISA, and 85.5% and 86.6% respectively, for rELISA. The value for the area under the ROC curve was 0.998 (0.994-1.0) (CI 95%) for eELISA and 0.929 (0.891-0.968) (CI 95%) for rELISA. The ROC curves for rLipL32 and sonicated antigen showed statistically significant differences (z = -3.826; p = 0.000). A three-way comparison showed statistically significant differences in the sensitivity and specificity of rELISA and eELISA. Our results showed that eELISA was more specific and sensitive than rELISA. The difference in performance (eELISA-rELISA) was 13.4% (4.03-23.28) (CI 95%) for sensitivity and 11.34 % (4.07-19.56) (CI 95%) for specificity. Our results show that the eELISA has a better diagnostic performance than rELISA for the detection of anti-Leptospira IgG antibodies in bovine serum.

In light optics, beams with orbital angular momentum (OAM) can be produced by employing a properly-tuned two-cylinder-lens arrangement, also called π/2 mode converter. It is not possible to convey this concept directly to the beam in an electron microscope due to the non-existence of cylinder lenses in commercial transmission electron microscopes (TEMs). A viable work-around are readily-available electron optical elements in the form of quadrupole lenses. In a proof-of-principle experiment in 2012, it has been shown that a single quadrupole in combination with a Hilbert phase-plate produces a spatially-confined, transient vortex mode. Here, an analogue to an optical π/2 mode converter is realized by repurposing a CEOS DCOR probe corrector in an aberration corrected TEM in a way that it resembles a dual cylinder lens using two quadrupoles. In order to verify the presence of OAM in the output beam, a fork dislocation grating is used as an OAM analyser. The possibility to use magnetic quadrupole fields instead of, e.g., prefabricated fork dislocation gratings to produce electron beams carrying OAM enhances the beam brightness by almost an order of magnitude and delivers switchable high-mode purity vortex beams without unwanted side-bands.

Resumen La leptospirosis continúa siendo hoy en día un problema para la salud pública, principalmente en poblaciones de bajos recursos socioeconómicos. En este trabajo se presenta la detección de leptospiras patógenas en muestras ambientales (aguas y barros) provenientes de regiones del norte argentino (provincias de Formosa, Salta, Santiago del Estero, Misiones y Chaco) con variadas características climatológicas habitadas por poblaciones vulnerables. De las 89 muestras analizadas, en el 24,7% fue posible detectar molecularmente la presencia de leptospiras patógenas. La prevalencia por tipo de muestra fue de 27,8% para las aguas y 11,8% para los barros. Todas las localidades muestreadas presentaron al menos una muestra positiva a alguna de las pruebas realizadas, por lo que el presente trabajo refleja la necesidad de profundizar los estudios de la leptospirosis en distintas regiones de la Argentina.


Abstract Leptospirosis remains as a major public health problem nowadays, mainly affecting vulnerable communities with low socioeconomic resources. In this study, the molecular detection of pathogenic leptospires from environmental samples (water and mud) from northern Argentina (Formosa, Salta, Santiago del Estero, Misiones and Chaco provinces) is described. Samples were obtained from regions with varied climatological features, all inhabited by vulnerable communities. From the 89 samples that were analyzed, 24.7% showed molecular evidence of the presence of pathogenic leptospires. Prevalence by sample type was: 27.8% in water samples and 11.8% in mud samples. All the sampled regions showed at least one positive sample. This result highlights the need of further research regarding leptospirosis in different regions of Argentina.

Abstract Molecular tools have improved conventional veterinary diagnosis. Acid nucleic extraction is a key step for downstream applications. This work aimed to compare the DNA extraction method Chelex-100 resin (M1) with Whatman® cards (M2), phenol-chloroform (M3), or commercial kits (M4), and to determine the most sensitive and inexpensive one for its diagnosis of animal pathogens that, despite their economic or zoonotic relevance, receive little attention. DNA was isolated from urine, organs, semen, blood and intestinal mucous, from the bacteria Leptospira interrogans serovar Pomona Pomona (by M1 and M2), Brucella melitensis (by M1, M3 and M4), and Salmonella ser. Abortusequi (by M1 and M4), and the parasites Leishmania spp. (by M1, M3 and M4), and Eimeria spp. (by M1 and M3), respectively. The sensitivity of each method was assayed by Polymerase Chain Reaction (PCR). The M1 showed similar sensitivity for Salmonella ser. Abortusequi, Leishmania spp., and Eimeria spp., being better for L. interrogans serovar Pomona Pomona and slightly lower for B. melitensis. For the first time, a simple and economic method was successfully employed for extracting DNA from these animal pathogens, especially important in low-resource settings, contributing to the diagnosis of leptospirosis, brucellosis, leishmaniasis, and coccidiosis; as well as to the molecular epidemiology of salmonellosis in stallion from semen samples.


Resumen Las técnicas moleculares han contribuido a mejorar el diagnóstico veterinario tradicional y la extracción de ácidos nucleicos es determinante. El objetivo de este trabajo fue comparar el método de extracción de ADN Chelex-100 (M1) con papel Whatman (M2), fenol-cloroformo (M3) o kits comerciales (M4), y determinar un método sensible y de bajo costo para el diagnóstico de patógenos de animales económica o zoonóticamente relevantes y que reciben poca atención. A partir de orina, órganos, semen, sangre y mucosa intestinal se extrajo el ADN de las bacterias Leptospira interrogans serovar Pomona Pomona (con M1 y M2), Brucella melitensis (con M1, M3 y M4), Salmonella ser. Abortusequi (M1 y M4), y de los parásitos Leishmania spp. (M1, M3 y M4) y Eimeria spp. (M1 y M3), respectivamente. La sensibilidad de los protocolos fue analizada por PCR. El método M1 demostró una sensibilidad similar para S. Abortusequi, Leishmania spp. y Eimeria spp., siendo mejor para L. interrogans y levemente menor para B. melitensis. Por primera vez se usó exitosamente en estos patógenos veterinarios un método simple y económico para extraer ADN, especialmente importante en laboratorios de bajos recursos económicos, contribuyendo al diagnóstico de leptospirosis, brucelosis, leishmaniasis y coccidiosis, así como también a la epidemiología molecular de salmonelosis en muestras de semen de caballos.

La leptospirosis bovina es una importante enfermedad zoonótica cuyo diagnóstico molecular está ampliamente divulgado. Sin embargo, no existe un método único de extracción de ADN para leptospiras patógenas a partir de muestras clínicas. En este trabajo se utilizó orina bovina contaminada con cultivo de L. interrogans serovar Pomona Pomona para analizar el mejor método comparando: M1.) resina Chelex-100, M2.) papel FTA Whatman y M3.) hervido de la muestra (protocolo casero). De estas tres técnicas, la primera (M1) presentó la mayor sensibilidad al realizar la PCR de diagnóstico, detectándose hasta 2x102 leptospiras/mL. La metodología aquí planteada resultó tener buen rendimiento para la detección de leptospiras en muestras clínicas animales, aunque es necesario su validación con mayor número y diversidad de muestras.


Bovine leptospirosis is an important zoonotic disease whose molecular diagnosis is widely reported. However, there is not a unique method of extraction of DNA for pathogenic leptospires using clinical samples. In this study, bovine urine was contaminated with pure culture of L. interrogans serovar Pomona Pomona in order to compare three of them: M1.) Chelex-100 resin, M2.) FTA Whatman paper and M3.) boiling of the sample (in-house protocol), being the first one the most sensitive when used in diagnostic PCR, detecting up to 2x102 leptospiras/mL. The methodology proposed in this study turned out to have good performance for the detection of leptospires in animal clinical samples, although it should be applied to a greater number of samples and in different stages of the pathology.

In optics, mode conversion is an elegant way to switch between Hermite Gaussian and Laguerre Gaussian beam profiles and thereby impart orbital angular momentum onto the beam and to create vortices. In optics such vortex beams can be produced in a setup consisting of two identical cylinder lenses. In electron optics, quadrupole lenses can be used for the same purpose. Here we investigate generalized asymmetric designs of a quadrupole mode converter that may be realized within the constraints of existing electron microscopes and can steer the development of dedicated vortex generators for high brilliance electron vortex probes of atomic scale.

PURPOSE: To describe the origin of the vessels supplying the anterior sub-axial cervical vertebrae (C3-C7) to further understand their potential influence on anterior bone loss after anterior cervical spinal surgery. METHOD: Cadaveric dissection was performed on ten adult human necks after latex perfusion of their subclavian, common carotid and vertebral arteries. The nutrient vessels of the sub-axial cervical spine were identified and traced to their origin. The course and distribution of these vessels and their nutrient foraminae are described. RESULTS: In all cases the anterior nutrient vessels were derived from the thyro-cervical trunk with branches that passed over the longus coli muscles forming a leash of vessels in the pre-vertebral fascia which subsequently extended in a frond-like pattern to pass onto the anterior aspect of vertebrae. The more cranial the cervical level the fewer the number of nutrient vessels and foraminae. The distribution of the foraminae on the anterior vertebral body followed the oblique supero-medial course of the nutrient vessels. CONCLUSION: Nutrient vessels perforate the cervical vertebrae on their anterior surface. These are derived from a leash of vessels that lie within the pre-vertebral fascia overlying the longus coli muscles. The origin of these vessels is the ascending cervical artery with a variable contribution from the transverse cervical artery.

BACKGROUND: The influence of personality on health-related quality of life in patients with multiple sclerosis has been the focus of previous studies showing that introversion and neuroticism were related with reduced health related quality of life. However, no data exist on the impact of temperament on quality of life in this patient group. METHODS: Between April 2014 and March 2016 139 multiple sclerosis patients were recruited from a specialized outpatient clinic of the general hospital of Vienna. Health-related quality of life was measured by "The Multiple Sclerosis International Quality of Life Questionnaire (MusiQol)", temperament by "Temperament Evaluation of Memphis, Pisa, Paris, and San Diego Questionnaire - Münster version" (briefTEMPS-M), and disability by the "Expanded disability status scale". All patients underwent a diagnostic psychiatric semi-structured interview (MINI). RESULTS: Known predictors (like disease duration, EDSS, psychiatric co-morbidities, immunomodulatory treatments) explain the proportion of variation in the outcome of MusiQol global index score in 30.9% in multi-variable linear regression analysis. It increased respectively to 40.3, 42.5, and 45.8% if adding the depressive, cyclothymic, or hyperthymic temperament to the list of variables. An increase of depressive and cyclothymic temperament scores significantly reduced global index score of MusiQol (p = 0.005, p = 0.002, respectively), while the hyperthymic temperament significantly raised it (p < 0.001). CONCLUSION: In MS patients, the depressive and cyclothymic temperament predict a lower and hyperthymic temperament an increased health-related quality of life, independent of current disability status, immunomodulatory treatments, and affective co-morbidities.

Interaction of the probe with the specimen in an electron microscope inevitably leads to entanglement between the probe and the scatterer. In spite of the importance of entanglement in many areas of modern physics, this subject has not been touched in the literature. Here, we develop some ideas about entanglement in electron microscopy for a number of scattering mechanisms. The relationship between entropy, density matrices, and coherence is discussed. In addition, we explore the questions "Why is Bragg scattering coherent and energy loss incoherent?" and "When does decoherence play a role?" It seems to be possible to measure decoherence on extremely short timescales of ∼10-8s. This is especially important in view of recent developments in ultrafast electron microscopy.

A strain characterization technique based on Moiré interferometry in a scanning transmission electron microscope (STEM) and geometrical phase analysis (GPA) method is demonstrated. The deformation field is first captured in a single STEM Moiré hologram composed of multiple sets of periodic fringes (Moiré patterns) generated from the interference between the periodic scanning grating, fixing the positions of the electron probe on the sample, and the crystal structure. Applying basic principles from sampling theory, the Moiré patterns arrangement is then simulated using a STEM electron micrograph reference to convert the experimental STEM Moiré hologram into information related to the crystal lattice periodicities. The GPA method is finally applied to extract the 2D relative strain and rotation fields. The STEM Moiré interferometry enables the local information to be de-magnified to a large length scale, comparable to what can be achieved in dark-field electron holography. The STEM Moiré GPA method thus extends the conventional high-resolution STEM GPA capabilities by providing comparable quantitative 2D strain mapping with a larger field of view (up to a few microns).

Energy-loss magnetic chiral dichroism (EMCD) is a versatile method for studying magnetic properties on the nanoscale. However, the classical EMCD technique is notorious for its low signal-to-noise ratio (SNR), which is why many experimentalists have adopted a convergent-beam approach. Here, we study the theoretical possibilities of using a convergent beam for EMCD. In particular, we study the influence of detector positioning as well as convergence and collection angles on the detectable EMCD signal. In addition, we analyse the expected SNR and give some guidelines for achieving optimal EMCD results.

We discuss the feasibility of detecting spin polarized electronic transitions with a vortex filter. This approach does not rely on the principal condition of the standard electron energy-loss magnetic chiral dichroism (EMCD) technique, the precise alignment of the crystal in order to use it as a beam splitter, and thus would pave the way for the application of EMCD to new classes of materials and problems, like amorphous magnetic alloys and interface magnetism. The dichroic signal strength at the L2, 3-edge of ferromagnetic Cobalt (Co) is estimated on theoretical grounds using a single atom scattering approach. To justify this approach, multi-slice simulations were carried out in order to confirm that orbital angular momentum (OAM) is conserved in amorphous materials over an extended range of sample thickness and also in very thin crystalline specimen, which is necessary for the detection of EMCD. Also artefact sources like spot size, mask tilt and astigmatism are discussed. In addition, the achievable SNR under typical experimental conditions is assessed.

A major challenge in the growing field of bioelectronic medicine is the development of tissue interface technologies promoting device integration with biological tissues. Materials based on organic bioelectronics show great promise due to a unique combination of electronic and ionic conductivity properties. In this review, we outline exciting developments in the field of organic bioelectronics and demonstrate the medical importance of these active, electronically controllable materials. Importantly, organic bioelectronics offer a means to control cell-surface attachment as required for many device-tissue applications. Experiments have shown that cells readily attach and proliferate on reduced but not oxidized organic bioelectronic materials. In another application, the active properties of organic bioelectronics were used to develop electronically triggered systems for drug release. After incorporating drugs by advanced loading strategies, small compound drugs were released upon electrochemical trigger, independent of charge. Another type of delivery device was used to achieve well-controlled, spatiotemporal delivery of cationic drugs. Via electrophoretic transport within a polymer, cations were delivered with single-cell precision. Finally, organic bioelectronic materials are commonly used as electrode coatings improving the electrical properties of recording and stimulation electrodes. Because such coatings drastically reduce the electrode impedance, smaller electrodes with improved signal-to-noise ratio can be fabricated. Thus, rapid technological advancement combined with the creation of tiny electronic devices reacting to changes in the tissue environment helps to promote the transition from standard pharmaceutical therapy to treatment based on 'electroceuticals'. Moreover, the widening repertoire of organic bioelectronics will expand the options for true biological interfaces, providing the basis for personalized bioelectronic medicine.

La leptospirosis es una zoonosis de amplia distribución mundial y de importancia en salud pública y veterinaria. El serogrupo Icterohaemorrhagiae toma importancia, ya que ha sido aislado con frecuencia en roedores y en casos clínicos de humanos, caninos, bovinos y porcinos; y se ha logrado aislar a partir de agua del rio Reconquista, provincia de Buenos Aires, Argentina. El objetivo de este estudio fue discriminar molecularmente entre las serovariedades Copenhageni e Icterohaemorrhagiae del serogrupo Icterohaemorrhagiae de importancia en Argentina, utilizando la técnica del análisis de repeticiones en tándem en múltiples locus (MLVA) y analizar las distancias génicas entre los genotipos determinados a partir de cepas aisladas de animales. Para ello, genotipificamos las 4 cepas referenciales del serogrupo Icterohaemorrhagiae de la especie Leptospira interrogans serovariedad Copenhageni cepa M20 y cepa Ictero I y de la serovariedad Copenhageni cepa RGA y cepa Fiocruz L1-130. Para este estudio se incluyeron 24 cepas aisladas de animales domésticos como silvestres. Logramos mediante esta técnica determinar un código numérico para cada serovariedad y así pudimos discriminar entre las serovariedades de este serogrupo. El análisis de coordenadas principales (PCoA) demostró la variabilidad génica existente entre las 4 cepas referenciales pertenecientes al serogrupo Icterohaemorrhagiae, el genotipo con mayor representación fue el serovar Copenhageni cepa Fiocruz L1-130 con un total de 13 cepas aisladas con ese perfil genético. Discriminar las serovariedades de este importante serogrupo nos permitirá en un futuro investigar si existen diferencias entre la sintomatología clínica y/o respuesta al tratamiento en los casos clínicos producidos por las distintas serovariedades del serogrupo Icterohaemorrhagiae discriminadas en este trabajo.


Leptospirosis is the world`s most widely distributed zoonosis and of great importance in animal health and in public health. The relevance of the serogroup Icterohaemorrhagiae relies on the frequent isolation from rodents and from clinical cases of humans, canines, bovines and swine; and this serogroup was also isolated from water samples of the Reconquista river, Buenos Aires province, Argentina. The objective of this study was to discriminate molecularly between serovars Copenhageni and Icterohaemorrhagiae being of importance in Argentina, using Multiple Locus Variable number tandem repeats analysis (MLVA) and analyzing the genetic distances among genotypes obtained from animal isolates in this study. We genotyped the four referential strains of the serogroup Icterohaemorrhagiae belonging to the species Leptospira interrogans serovar Copenhageni strain M20 and strain Ictero I and of the serovar Copenhageni strain RGA and strain Fiocruz L1-130. In this study we included 24 strains isolated from animals domestic and wildlife. Using this technique we could determine numeric codes for each serovar and in this way, discriminate between serovars of this serogroup. The principal coordinates analysis (PCoA) showed the genetic variability of the four referential strains from the serogroup Icterohaemorrhagiae, the genotype with better representation was serovar Copenhageni strain Fiocruz L1-130 with 13 isolated strains with this genetic profile. Discrimination of the serovars from this important serogroup allows to establish differences between clinical signs and/or response to treatment in the clinical cases produced by different serovars of the serogroup Icterohaemorrhagiae successfully distinguished in this study.

BACKGROUND: The removal of well-fixed acetabular components following THA (total hip arthroplasty) is a difficult operation and could be accompanied by the loss of acetabular bone stock. The optimal method for fixation is still under debate. The aim of this pilot study was to compare the tear-out resistance and failure behavior between osseo-integrated and non-integrated screw cups. Furthermore, we examined whether there are differences in the properties mentioned between screw sockets and cemented polyethylene cups. HYPOTHESIS: Tear-out resistance and related mechanical work required for the tear-out of osseo-integrated screw sockets are higher than in non-integrated screw sockets. PATIENTS AND METHODS: Ten human coxal bones from six cadavers with osseo-integrated screw sockets (n=4), non-integrated (implanted post-mortem, n=3) screw sockets and cemented polyethylene cups (n=3) were used for tear-out testing. The parameters axial failure load and mechanical work for tear-out were introduced as measures for determining the stability of acetabular components following THA. RESULTS: The osseo-integrated screw sockets yielded slightly higher tear-out resistance (1.61±0.26kN) and related mechanical work compared to the non-integrated screw sockets (1.23±0.39kN, P=0.4). The cemented polyethylene cups yielded the lowest tear-out resistance with a failure load of 1.18±0.24kN. Compared to the screw cups implanted while alive, they also differ on a non-significant level (P=0.1). Osseous failure patterns differed especially for the screw sockets compared to the cemented polyethylene cups. DISCUSSION: Osseo-integration did not greatly influence the tear-out stability in cementless screw sockets following axial loading. Furthermore, the strength of the bone-implant-interface of cementless screw sockets appears to be similar to cemented polyethylene cups. However, given the high failure load, high mechanical load and because of the related bone failure patterns, removal should not be performed by means of tear-out but rather by osteotomes or other curved cutting devices to preserve the acetabular bone stock. Standard electron optics predicts Larmor image rotation in the magnetic lens field of a TEM. Introducing the possibility to produce electron vortex beams with quantized orbital angular momentum brought up the question of their rotational dynamics in the presence of a magnetic field. Recently, it has been shown that electron vortex beams can be prepared as free electron Landau states showing peculiar rotational dynamics, including no and cyclotron (double-Larmor) rotation. Additionally very fast Gouy rotation of electron vortex beams has been observed. In this work a model is developed which reveals that the rotational dynamics of electron vortices are a combination of slow Larmor and fast Gouy rotations and that the Landau states naturally occur in the transition region in between the two regimes. This more general picture is confirmed by experimental data showing an extended set of peculiar rotations, including no, cyclotron, Larmor and rapid Gouy rotations all present in one single convergent electron vortex beam.

To facilitate automated real-time monitoring of the formation or disruption of an epithelial monolayer, phase angle spectroscopy was performed in custom-designed cell culture dishes including a transparent conductive polymer-based 2-electrode device. A peak in φ amplitude at 103-105 Hz specifically correlated to increased capacitance by proliferating cells. Dynamic monitoring of cell density, and barrier integrity during monolayer formation and disruption were investigated in combination with live-cell microscopy.

Landau levels and states of electrons in a magnetic field are fundamental quantum entities underlying the quantum Hall and related effects in condensed matter physics. However, the real-space properties and observation of Landau wave functions remain elusive. Here we report the real-space observation of Landau states and the internal rotational dynamics of free electrons. States with different quantum numbers are produced using nanometre-sized electron vortex beams, with a radius chosen to match the waist of the Landau states, in a quasi-uniform magnetic field. Scanning the beams along the propagation direction, we reconstruct the rotational dynamics of the Landau wave functions with angular frequency ~100 GHz. We observe that Landau modes with different azimuthal quantum numbers belong to three classes, which are characterized by rotations with zero, Larmor and cyclotron frequencies, respectively. This is in sharp contrast to the uniform cyclotron rotation of classical electrons, and in perfect agreement with recent theoretical predictions.
